| text | HHV8 immediate/early genes are transcribed by host RNA polymerase II holoenzyme complex (POLR2), and their mRNAs are 5′-capped and 3′-polyadenylated by the standard cellular co-transcriptional machinery — KSHV does not encode its own capping enzyme for these transcripts (Purushothaman et al., 2015; for a recent review see Losay & Damiana, 2025). Roughly 30% of Kaposi's sarcoma-associated herpesvirus (KSHV/HHV-8) genes are expressed as spliced transcripts, with the host spliceosome removing canonical GU-AG introns from immediate-early and early pre-mRNAs such as ORF50 and K8 (Majerciak et al. 2022). The viral RNA-binding protein ORF57 acts as a positive regulator, promoting splicing of several immediate-early and early transcripts, in part by binding the cellular splicing factor SRSF3 to relieve its suppressive activity; genome-wide RNA-seq of patient-derived BCBL-1 primary effusion lymphoma cells carrying wild-type versus ORF57-null genomes mapped these splicing events (Majerciak et al. 2022). The bicistronic ORF70/K3 transcript yields multiple alternatively spliced isoforms, including a cycloheximide-resistant doubly spliced immediate-early RNA, indicating that splicing occurs at the earliest stage of reactivation (Rimessi et al. 2001; Taylor et al. 2005). |
