textORF57 bound to HHV8 intronless mRNA engages the export adaptor ALYREF, and ALYREF in turn recruits the DEAD-box RNA helicase DDX39B, extending the viral ribonucleoprotein towards a complete hTREX assembly. The direct ALYREF–DDX39B contact was mapped by GST pull-downs to the C-terminus of ALYREF, with full-length DDX39B or its C-terminal half binding ALYREF from nuclear extract (Masuda et al. 2005). Reconstitution with purified recombinant proteins showed that ORF57 does not associate with DDX39B on its own but does so when ALYREF is added, demonstrating that ALYREF bridges DDX39B to ORF57 (Jackson et al. 2011), and co-immunoprecipitations from ORF57-expressing and HHV8-infected cells confirmed that DDX39B is recruited together with ALYREF onto viral intronless transcripts in an ORF57-dependent manner (Boyne et al. 2008; Jackson et al. 2011).
