| text | On the HHV8 intronless viral mRNA, DDX39B — already tethered through ORF57 and ALYREF — binds the THO complex via THOC1 (HPR1) to complete assembly of the full TREX export complex on the transcript (Boyne et al. 2008). ATP hydrolysis by DDX39B is required for this remodelling step (Schumann et al. 2016). Reconstitutive GST pull-downs showed that GST-THOC1 captured ORF57 only when both ALYREF and purified DDX39B were present, identifying DDX39B as the bridge between the ALYREF-bound viral ribonucleoprotein and the THO complex (Boyne et al. 2008), consistent with the direct ALYREF–DDX39B interaction characterised in uninfected cells (Luo et al. 2001). RNA-immunoprecipitation from cells expressing intronless HHV8 ORF47 or gB mRNAs confirmed that THOC1, alongside DDX39B and ALYREF, is recruited to these transcripts only in the presence of ORF57 (Boyne et al. 2008), and depletion of DDX39B abolishes ORF57-mediated assembly of the export-competent vRNP and viral intronless mRNA export (Jackson et al. 2011; Majerciak et al. 2010). |
