| text | Within the viral messenger ribonucleoprotein, the TREX-associated adaptor ALYREF engages the NXF1,2:NXT1 (TAP:p15) heterodimer through direct protein–protein contact, thereby loading the bulk mRNA export receptor onto the ORF57:TREX/CBC-bound HHV8 transcript. Biochemical mapping placed the ALYREF docking site on the N-terminal and leucine-rich-repeat domains of NXF1,2, a surface distinct from the Ntf2-like (M) domain that contacts THOC5, so that ALYREF and the THO subunit can occupy NXF1,2:NXT1 simultaneously in a hetero-tetrameric complex assembled in GST pull-downs with recombinant proteins (Katahira et al. 2009). GST pull-down of in vitro-translated ALYREF by recombinant NXF1(1–231) further confirmed that the ALYREF–NXF1 interaction is direct and is competed by DDX39B, indicating that release of the helicase precedes or accompanies NXF1,2 recruitment (Taniguchi and Ohno, 2008). |
