| text | Within the HHV8 lytic replication machinery, the ORF56 primase subunit, in association with the HELI helicase and the ORF40/41 primase-associated factor, synthesizes short RNA primers on the DBP-coated single-stranded DNA template, consuming NTPs and releasing pyrophosphate (PPi). ORF56, HELI and ORF40/41 were shown to be strictly required for HHV8 oriLyt-dependent DNA amplification in cotransfection–replication assays, since omission of any one of these genes abolished replication (AuCoin et al. 2004; Wu et al. 2001). Details of the process are unknown. |
