| text | The HHV8 bZIP protein K8 directly binds the viral noncoding RNA T1.4, which is transcribed from the ori-Lyt (L) locus, with CLIP-seq of TPA-induced BCBL-1 cells identifying T1.4 as one of three major viral RNAs cross-linked to K8 and in vitro pulldowns using biotinylated T1.4 and bacterially expressed GST-K8 confirming a direct, specific interaction (Liu et al. 2018). Truncation analysis localised the contacts to both the 5′ GC-repeat and the 3′ non-repeat regions of T1.4, while mutagenesis of K8 mapped the RNA-binding activity to a GRYGR motif (residues 173–177) within the basic domain, since deletion of these residues or substitution to GDDGR abolished T1.4 binding (Liu et al. 2018). |
