textAfter translation starts, the signal peptide or signal anchor of the nascent polypeptide protrudes from the exit tunnel of the ribosome and is bound by the SRP54 subunit of the signal recognition particle (SRP) (Lakkaraju et al. 2008, Voorhees and Hegde 2015, Lee et al. 2018, Lee et al. 2021, inferred from canine homologs in Walter et al. 1981, Halic et al. 2006, reviewed in Nagai et al. 2003, Akopian et al. 2013). A signal peptide is a short (~20 amino acid residue) hydrophobic region at the N-terminus of an endoplasmic reticulum-targeted (ER-targeted) protein that is cleaved during the translocation of the protein across the ER membrane. A signal anchor is also a hydrophobic region, however it is not cleaved and remains embedded as a transmembrane domain in the ER membrane after translocation. The binding of the SRP arrests translation elongation (inferred from canine homologs in Walter and Blobel 1981).
