textThree HHV8 (KSHV) genes are transcribed from the LANA promoter as a polycistronic pre-mRNA: ORF73 (encoding LANA1), ORF72 (encoding vCyclin) and ORF71 (encoding vFLIP) (Dittmer et al. 1998; Talbot et al. 1999; Sarid et al. 1999). Transcription of these genes is essential for the establishment of latent infection, but they are also transcribed during lytic infection, with activating transcription factors differing between latent and lytic stages of the HHV8 life cycle (Dittmer et al. 1998; Talbot et al. 1999; Sarid et al. 1999). Introns separating ORF73, ORF72 and ORF71 portions of the polycistronic pre-mRNA are subsequently alternatively spliced, producing several mature transcripts with overlapping 3' ends that are either tricistronic or bicistronic (Dittmer et al. 1998; Talbot et al. 1999; Sarid et al. 1999). HHV8-encoded microRNA, miR-K10, negatively regulates translation of all transcripts derived from the polycistronic ORF73-ORF72-ORF71 pre-mRNA (reviewed in Zhu et al. 2014). Prostaglandin E2 (PGE2) positively regulates transcription from the LANA promoter and the establishment of latent infection (George Paul et al. 2010).
