| text | Na+/I- (sodium/iodide) symporter (NIS, SLC5A5) binds Transitional endoplasmic reticulum ATPase VCP (aka valosin-containing protein) (Fletcher et al., 2019). VCP is part of endoplasmic reticulum (ER) quality control: a complex containing VCP deglycosylates and unfolds target proteins, and reroutes them to the cytosol for proteasomal degradation. Ebastine is an approved H1 antihistamine and clotrimazole is an approved antifungal. Both act as VCP inhibitors, abolishing the repression of SLC5A5 maturation by VCP in human and mouse thyroid and breast cancer models (Fletcher et al., 2019; reviewed in Riesco-Eizaguirre et al., 2021). |
