textNa/K-ATPase (NKA) complex pumps 3 Na+ ions to extracellular space in exchange for 2 K+ ions, using up one ATP molecule (Guo et al., 2022; Kanai et al., 2023; reviewed in Contreras et al., 2024). This creates a sodium concentration imbalance in the thyroid cell that drives iodide import via NIS (see, e.g., Paire et al., 1998). In rat thyroid cells, mRNA for ATP1A1, ATP1A2, and ATP1A3 alpha subunits has been detected (Pekary et al., 1997), while in pig thyroid cells, expression of the ATP1A1 subunit has been confirmed (Paire et al., 1998). Human ATP1A1 subunit has been shown to associate with the regulatory subunit FXYD3 when co-expressed in Xenopus oocytes (Bibert et al., 2006).
