| text | GMNC is a coiled-coil-containing transcriptional regulator that plays a key role in the generation of multiciliated cells (MCCs) through its regulation of expression of other key target genes including MCIDAS and TP73 (Lu et al, 2019; Terre et al, 2016; Arbi et al, 2016; reviewed in Cumplido-Laso et al, 2023; Lewis and Stracker, 2021; Arbi et al, 2018). GMNC expression is negatively regulated by NOTCH signaling (Kyrousi et al, 2015; Arbi et al, 2016; Zhou et al, 2015; Terre et al, 2016). GMNC and MCIDAS share overlapping but non-redundant roles in the formation of multiciliated cells: GMNC is required to specify the MCC fate through upregulation of genes such as TP73, FOXJ1, RFX family members and MCIDAS, whereas MCIDAS is additionally essential for upregulating genes involved in basal body amplification that is required for the formation of multiple cilia (Lu et al, 2019; Lewis et al, 2023; reviewed in Cumplido-Laso et al, 2023; Lewis and Stracker, 2021). Consistent with this, expression of MCIDAS is compromised in GMNC mutant mice (Terre et al, 2016; Lu et al, 2019). |
