| text | GMNC is a coiled-coil containing protein in the MCIDAS, GMNN (Geminin) family (reviewed in Arbi et al, 2018; Lewis and Stracker, 2021). Like MCIDAS, GMNC is a key player in the transcriptional regulation of multiciliogenesis (Stubbs et al, 2012; Zhou et al, 2015; Arbi et al, 2016; Terre et al, 2016; Lu et al, 2019). Lacking a DNA-binding domain, GMNC is recruited to target genes as part of a ternary complex involving TFDP1 and E2F4 or E2F5 (Lu et al, 2019; Chong et al, 2018; reviewed in Lewis and Stracker, 2021; Cumplido-Laso et al, 2023). Note that although this reaction shows GMNC binding to TFDP1 and E2F transcription factors as a dimer, this has not been experimentally demonstrated. Analysis of the roles for GMNC and MCIDAS in initiating formation of multiciliated cells (MCCs) suggests that both contribute to the upregulation of targets such as TP73, FOXJ1 and RFX proteins, but that MCIDAS is exclusively responsible for the upregulation of targets controlling basal body amplification such as DEUP1, CCNO, CCDC78 and CDC20B (Lu et al, 2019; Lewis et al, 2023; reviewed in Lewis and Stracker, 2021; Cumplido-Laso et al, 2023). |
