| text | Host ribosomes translate DENV genomic RNA, producing a polyprotein of 3,391 aa length and mass of about 380 kDa. The short 5′ untranslated region (5′-UTR) of DENV genomic RNA has been shown to function as an internal ribosomal entry site (IRES) in BHK (hamster) cells (Song et al., 2019), and its activity is enhanced in human HEK293T cells when cap-dependent translation initiation is inhibited (Fernandez-Garcia et al., 2021). This cap-independent, IRES-mediated translation is an alternative mechanism of DENV mRNA translation (Song et al., 2019; Fernandez-Garcia et al., 2021). While DENV can also utilize a non-IRES cap-independent mechanism when eIF4E is inhibited in BHK and Vero (monkey) cells (Edgil et al., 2006), its IRES-dependent mechanism is notably active under conditions of impaired canonical translation, such as eIF4G cleavage (Fernandez-Garcia et al., 2021; Fernandez-Garcia et al., 2024) and eIF2A phosphorylation (Ricciardi-Jorge et al., 2023). However, Hodge et al. (2019) argue against IRES-mediated translation in flaviviruses, noting that canonical IRES-like RNA structures have not been identified in DENV genomes. |
