textMCIDAS is a coiled-coil-containing transcriptional regulator that plays a key role in the generation of multiciliated cells through its regulation of expression of other key target genes including TP73 (Stubbs et al, 2012; Boon and Wallingford, 2014; Nemajerova et al, 2016; reviewed in Nemajerova et al, 2018; Cumplido-Laso et al, 2023). In addition to regulating multiciliation-specific transcriptional events, MCIDAS has more recently also been show to translocate to the cytosol where it contributes to centriole biogenesis (Lu et al, 2025).
Expression of MCIDAS is regulated by the binding of GRHL2 to cognate sites in its promoter, as assessed by ChIP in human bronchial epithelial cells (Gao et al, 2013; Gao et al, 2015). MCIDAS expression is additionally negatively regulated by NOTCH signaling (Stubbs et al, 2012). Expression of MCIDAS also appears to depend on GMNC, a related coiled-coil protein that has distinct but overlapping roles in multiciliogenesis (Lu et al, 2019). GMNC is required to specify the multiciliated cell (MCC) fate through upregulation of genes such as FOXJ1, RFX family members and MCIDAS, whereas MCIDAS is essential for upregulating genes involved in basal body amplification that is required for the formation of multiple cilia (Lu et al, 2019); reviewed in Lewis et al, 2023; Cumplido-Laso et al, 2023). Consistent with this, expression of Mcidas is compromised in Gmnc mutant mice (Terre et al, 2016; Lu et al, 2019).
