textA recombinant mouse CBLL1 (Hakai), an E3 ubiquitin ligase, was first shown to ubiquitinate a recombinant mouse CDH1 (E-cadherin) in human HEK293 cell line and this ubiquitination was dependent on the expression of a recombinant avian SRC (Fujita et al. 2002). Ubiquitination of the endogenous canine CDH1 in canine cell line MDCK is stimulated by ectopically overexpressed recombinant moues CBLL1 and further enhanced by treatment with HGF, a ligand of receptor tyrosine kinases EGFR and MET, which can also activate SRC (Fujita et al. 2002). In the human breast cancer cell line MCF7, ubiquitination of CDH1 downstream of calcium depletion-activated CDC42 RHO GTPase and SRC kinase is dependent on CBLL1 (Shen et al. 2008). In the human hepatocellular carcinoma cell line HepG2, activation of SRC and FYN kinases increases both interaction between CBLL1 and CDH1 and the level of ubiquitinated CDH1 (Chen et al. 2009). The integrin-interacting protein BSG (also known as Basigin or CD147), which triggers SRC activation in HepG2 cells, promotes CBLL1-mediated ubiquitination of CDH1 (Lu et al. 2018). In a study in which recombinant mouse CDH1 constructs, either full-length or just involving the juxtamembrane domain (JMD) of CDH1, were expressed in the canine MDCK cell line, it was found that two conserved lysine residues in the JMD (K740 and K818 in mouse CDH1, corresponding to K738 and K816 in human CDH1) are required for CDH1 ubiquitination and that CDH1 ubiquitination interferes with CTNND1 (p120 catenin or delta-catenin) binding (Hartsock and Nelson 2012). In the human colon carcinoma cell line T84, IFNG (inteferfon-gamma)-stimulated ubiquitination of CDH1 is dependent on CBLL1 (Smyth et al. 2012). Using recombinant human proteins expressed in human colon adenocarcinoma cell line Caco-2, it was found that the kinesin protein KIFC3 can recruit a deubiquitinating enzyme USP47 to adherens junctions to remove ubiquitins ligated to CDH1 by CBLL1 and prevent CDH1 degradation (Sako-Kubota et al. 2014).
