| text | MITF-M binds to 14-3-3 proteins in the cytosol, sequestering the transcription factor away from target genes. Binding to 14-3-3 family members depends on residues 141-191 of MITF-M and is enhanced by MARK3 (also known as CTAK1)-dependent phosphorylation at serine 173 (Bronisz et al, 2006). MARK3 binding and phosphorylation and 14-3-3 binding is inhibited in the presence of cytokines CSF (colony stimulating factor) and TNFSF11 (Tumor necrosis factor ligand superfamily member 11; also known as RANKL, receptor activator of nuclear factor-kappa beta ligand), both known to induce osteoclast differentiation (Bronisz et al, 2006 reviewed in Boyle et al, 2003). |
