textIn the retinal vasculature of mice, activation of the Dll4 promoter is induced by Vegfa acting via its receptor Vegfr2 (Lobov et al. 2007).
MEF2 transcription factors activate the expression of critical sprouting angiogenesis regulators including DLL4, activated by VEGFA signaling (Sacilotto et al. 2016). VEGFA signaling induces the release of repressive histone deacetylases and recruitment of the histone acetyltransferase, EP300, to regulatory elements of MEF2 target genes, including DLL4 (Sacilotto et al. 2016).
In response to VEGFA or Angiopoietin, phosphorylated ERG binds enhancers in the DLL4 gene located upstream of the transcription start site and within the third intron and activates transcription (Fish et al. 2017, Shah et al. 2017, Kalna et al. 2019, inferred from mouse homologs in Wythe et al. 2013).
