| text | MLL1 (Mixed-Lineage Leukemia 1) is a protein that functions as a histone methyltransferase. It plays a crucial role in regulating gene expression by modifying chromatin structure. MLL-1 specifically methylates histone H3 at lysine 4 (H3K4), which is generally associated with active transcription. MLL-1 mediates CD274 (PDL-1_ promoter trimethylation (H3K4me3) and increases PDL1 expression in human and mouse pancreatic cancer cells. Chromatin immunoprecipitation (ChIP) analysis revealed H3K4me3 levels are enriched in the proximal CD274 promoter region in both PANC02-H7 and UN-KC-6141 orthotopic tumors, whereas a minimal level of H3K4me3 was detected in the cd274 promoter region in normal pancreas in vivo (Lu et al.,2017). MLL3 (Mixed-Lineage Leukemia 1, also known as KMT2C) belong to the MLL/COMPASS family, and have been identified as the major H3K4 mono-methyltransferase for enhancer. In PC3 human prostate cancer cell line, MLL3 bound in the enhancer region and the promoter region of PD-L1. Quantitative reverse transcription polymerase chain reaction (RT-qPCR) results showed that knocking down MLL3, but not the other MLLs, reduced the RNA levels of PD-L1. MLL3 regulated PD-L1 at the level of transcription by increasing H3K4 mono-methylation in the enhancer region (Xiong et al., 2019) |
