Query author contributions in Reactome
Reactome depends on collaboration between our curation team and outside experts to assemble and peer-review its pathway modules. The integration of ORCID within Reactome enables us to meet a key challenge with authoring, curating and reviewing biological information by incentivizing and crediting the external experts that contribute their expertise and time to the Reactome curation process. More information is available at ORCID and Reactome.
If you have an ORCID ID that is not listed on this page, please forward this information to us and we will update your Reactome pathway records.

| comment | FUNCTION Receptor involved in axon guidance during development. Acts as a multifunctional regulator of pathfinding that simultaneously mediates NELL2 repulsion, inhibits SLIT repulsion, and facilitates Netrin-1/NTN1 attraction (PubMed:15084255, PubMed:18466743, PubMed:25433640, PubMed:30843071). In spinal cord development plays a role in guiding commissural axons probably by preventing premature sensitivity to Slit proteins thus inhibiting Slit signaling through ROBO1/ROBO2 (PubMed:15084255, PubMed:18466743). Binding OF NELL2 to the receptor ROBO3 promotes oligomerization of ROBO3, resulting in the repulsion of commissural axons in the midline (PubMed:30843071, PubMed:32198364). ROBO3 also indirectly boosts axon attraction to NTN1 without interacting with NTN1 itself (PubMed:25433640).FUNCTION Mediates NELL2 premature repulsion of commissural axons during midline crossing.FUNCTION After midline crossing by the commissural axons, may, in concert with ROBO1 and ROBO2, prevent midline recrossing. Does not mediate NELL2 signaling.SUBUNIT Interacts (via Fibronectin type-III 1 domain) with NELL2 (via the EGF domains) with a 3:3 stoichiometry; this interaction promotes oligomerization of ROBO3 resulting in the repulsion of commissural axons in the midline.SUBCELLULAR LOCATION A number of isoforms are produced.TISSUE SPECIFICITY Detected in embryonal spinal cord and hindbrain.DEVELOPMENTAL STAGE At 11.5 days post-conception (dpc) highly expressed in spinal cord commissural axons before crossing the floor plate (at protein level) (PubMed:14678835, PubMed:18466743). At 11.5 and 12.5 dpc, expressed in the midline and in the postcrossing ventral fiber tracts (at protein level) (PubMed:18466743). At 10.5 dpc is expressed in long stripes in the dorso-ventral part of the neural tube (PubMed:14678835). Later at 11.5, 12.5 and 13.5 dpc expression gradually moves dorsally and medially towards the roof plate (PubMed:14678835). After midline crossing initially continues to be expressed but then gets down-regulated (PubMed:14678835).DEVELOPMENTAL STAGE At 11.5 and 12.5 dpc, highly expressed on commissural axons before and during midline crossing (at protein level).DEVELOPMENTAL STAGE At 11.5 dpc expressed at a low level on the portion of commissural axons distal to the midline and a higher level in the same region at 12.5 dpc (at protein level).DISRUPTION PHENOTYPE Deficient mice lack embryonal spinal cord commissural axons crossing the floor plate.SIMILARITY Belongs to the immunoglobulin superfamily. ROBO family. |
