textThe MIR211 gene encoding mircroRNA miR-211 lies in intron 6 of the MITF-M target gene TRPM1, and as such is a co-transcriptional target of MITF-M-mediated regulation (Hunter et al, 1998). miR-211 expression is restricted to the melanocyte lineage, and its expression is frequently downregulated during melanoma progression (Guar et al, 2007; Duncan et al, 1998; Duncan et al, 2001). Direct binding and activation of MITF-M to the TRPM1/miR-211 locus has been demonstrated by ChIP, EMSA and reporter gene assay (Miller et al, 2004; Miller et al, 2005; Strub et al, 2011).
MITF-M-dependent miR-211 expression may contribute to melanocyte development and progression of melanoma through regulation of targets such as POU3F2, SERPINC3, AP1S2, IGFBP5 and SOX11, among others, whose disruption affects invasive properties of cells (Margue et al, 2013; Boyle et al, 2011; Levy et al, 2010; De Luca et al, 2016; reviewed in Goding and Arnheiter, 2019).
