| text | TRPM1 encodes a cation channel that transfers cations into the cytoplasm resulting in membrane depolarization. TRPM1 contains three E-boxes in its promoter that are bound by MITF-M to activate transcription, as assessed by ChIP, EMSA and reporter gene assay (Miller et al, 2004; Miller et al, 2005; Strub et al, 2011). Interestingly, non-sumoylatable MITF-M proteins with mutations at lysine residues 182 and 316 show a higher activity on a TRPM1 promoter construct provided it contains 3 E-boxes, but not when it contains only one E-box (Miller et al, 2005). The gene for miR-211 is contained in an intron of TRPM1, making the miRNA a co-transcriptional target of MITF-M (Levy et al, 2010; reviewed in Goding and Arnheiter, 2019). |
