| text | MITF-M-dependent transcriptional targets RAB27A and MYO5A play a role in melanosome organization and maturation by regulating melanosome distribution (Chiaverini et al, 2008; Alves et al, 2017; reviewed in Hume and Seabra, 2011; Seabra and Coudrier, 2004; Cheli et al, 2010; Goding and Arnheiter, 2019). Melanosome-bound RAB27A and the actin-binding protein MYO5A form a complex mediated by the RAB effector protein MLPH, also known as melanophilin, or the RAB effector MYRIP (Nagashima et al, 2002; El-Amraoui et al, 2002). This heteromeric complex directs localization of melanosomes to the tips of dendrites prior to loading of melanosomes into adjacent keratinocytes (Bahadoran et al, 2001; Nagashima et al, 2002; El-Amraoui et al, 2002; Fukuda et al, 2002; Alves et al, 2013; Evans et al, 2014; Alves et al, 2017). Docking at the plasma membrane may be facilitated by SYTL2, another RAB27A-binding protein (Kuroda and Fukuda, 2004; Menasche et al, 2008; Yu et al, 2007. Note that the existence of a complex containing each of SYTL2, RAB27A, MYO5A and either MLPH or MYRIP has not been explicitly demonstrated. |
