textThe JAK family member TYK2 is phosphorylated in anaplastic large cell lymphoma (ALCL) in both NPM1-ALK positive and ALK- cases. TYK2 phosphorylation promotes phosphorylation of both STAT1 and STAT3, although direct TYK2-mediated phosphorylation has not been demonstrated. Active TYK2 additionally contributes to increased expression of the pro-survival gene MCL1,. Consistent with this, knockdown of TYK2 induces death in human ALCL cell lines (Prutsch et al, 2019).
