textPrior to gastrulation, Sox2 is expressed throughout the epiblast (Wood and Episkopou 1999). During gastrulation Sox2 becomes restricted to the presumptive neuroectoderm in the anterior ectoderm and persists in the neural plate and neural tube (Wood and Episkopou 1999, Avilion et al. 2003, Uchikawa et al. 2011, Iwafuchi-Doi et al. 2012). Sox2 is considered to be the earliest marker of developing neurectoderm (Tang et al. 2015). In the anterior neural plate, first Pou5f1, then Pou3f1 bind the N2 enhancer upstream of the Sox2 coding region and activate transcription of Sox2 (Iwafuchi Doi et al. 2011, Zhu et al. 2014). In posterior neural plate, Wnt signaling and Fgf signaling act on the N1 enhancer downstream of the Sox2 coding region (Takemoto et al. 2006). Forced expression of Sox2 is able to direct embryonic stem cells to differentiate into neuroectoderm (Zhao et al. 2004).
