textIn mouse embryos, Sox17 is first expressed in the visceral endoderm, then in presumptive definitive endoderm before and during intercalation with visceral endoderm (Viotti et al. 2014). Expression of SOX17 is activated by WNT signaling via LEF1:CTNNB1 binding the SOX17 promoter (inferred from mouse homologs). During differentiation of human pluripotent stem cells to endoderm, expression of SOX17 is activated by NODAL signaling via phosphorylated SMAD2 and SMAD3 (Brown et al. 2011, Teo et al. 2011). GATA6, EOMES (Chia et al. 2019), BMP signaling (Xu et al. 2011), and MIXL1 (inferred from mouse homologs) also regulate SOX17.
