textNODAL signaling in the anterior primitive streak leads to the phosphorylation of SMAD2 and SMAD3, which co-translocate with SMAD4 to the nucleus and bind the promoter of the GATA4 gene (inferred from human pluripotent stem cells in Brown et al. 2011). GATA6 and EOMES (Chia et al. 2019) and TBXT (Faial et al. 2015), initially expressed in the primitive streak, also bind and activate transcription of GATA4. FOXA2 binds an intronic enhancer in the GATA4 gene in definitive endoderm progenitors and activates transcription in the gut tube populated by the endoderm (inferred from mouse homologs).
