textThe SET1A complex binds to HCFC1 (also known as Host cell factor 1 or HCF-1) (Wysocka et al. 2003; Tyagi et al. 2007; Lee and Skalnik 2008). The interaction involves the N-terminal half of HCFC1 and the evolutionarily conserved HCF-1 binding motif EHNY in the C-terminal half of SETD1A (Wysocka et al. 2003), which is not present in SETD1B (Lee et al. 2007). HCFC1 may enable formation of super-complexes that involve the SET1A complex and the SIN3A histone deacetylase complex (HDAC) (Wysocka et al. 2003). HCFC1 may enable association of the SET1A complex with E2F proteins, leading to the recruitment of SET1A complexes to promoters of E2F-target genes (Tyagi et al. 2007). Antibodies against WDR82, a protein that binds to the core SET1A complex, can pull down HCFC1, so HCFC1 is shown to bind to the SET1A complex or the SET1A complex associated with CXXC1 and WDR82 (Wu et al. 2008; van Nuland et al. 2013).
