textSET1A complex associates with CXXC1 (also known as CFP1) and WDR82 proteins, which interact with the C-terminus and the N-terminus of SETD1A, respectively (Lee et al. 2007; Lee and Skalnik 2008; Wu et al. 2008; van Nuland et al. 2013). The association of CXXC1 and the WRAD complex with the C-terminus of SETD1A increases the protein stability of SETD1A (Lee et al. 2007; Wu et al. 2008). WDR82 binds to RNA polymerase II phosphorylated on serine S5 of heptad repeats during transcription initiation and may act to recruit the SET1A complex to promoters of transcriptionally active genes (Lee and Skalnik 2008). The association of WDR82 with chromatin likely requires prior ubiquitination of histone H2B (Wu et al. 2008). WDR82 may facilitate association of CXXC1 with SETD1A but does not affect the association of SETD1A with the WRAD complex (Lee and Skalnik 2008). Based on quantitative analysis in HeLa cells, CXXC1 may bind the SET1A complex as a dimer (van Nuland et al. 2013).
