textTranscription factors CDX4 and MEN1 are both needed for HOXA9 expression and normal hematopoiesis. In mouse bone marrow cell lines transformed by the overexpression of the human leukemia-derived KMT2A (MLL1) fusion protein MLL-AF9, CDX4 and MEN1 were shown to directly stimulate HOXA9 gene transcription, where the establishment of H3K4me3 epigenetic marks of transcriptionally active chromatin by the complex of MLL1 and MEN1 likely enables access of CDX4 to the HOXA9 gene promoter (Yan et al. 2006). In the U937 promonocytic cancer cell line derived from histiocytic lymphoma, the leukemia-derived fusion protein of KMT2A (MLL1), MLL-ELL, increases ITGB3 transcription in CDX4- HOXA9- and HOXA10-dependent manner (Shah et al. 2016). CDX4-mediated upregulation of hematopoiesis-related HOX family genes is evolutionarily conserved. The cdx4 gene is mutated in kugelig (kgg), a zebrafish mutant with an early defect in haematopoiesis and aberrant expression of hox genes that can be rescued by overexpression of hoxb7a and hoxa9a (Davidson et al. 2003). Human CDX4 directly stimulates transcription of the human HOXA10 gene. HOXA10 is also able to bind to the CDX4 gene promoter elements and activate CDX4 transcription, thus creating a positive feedback loop (Bei et al. 2011).
