| text | Transcription factors CDX4 and MEN1 are both needed for HOXA9 expression and normal hematopoiesis. By chromatin immunoprecipitation (ChIP) in mouse bone marrow cell lines transformed by the overexpression of the human leukemia-derived KMT2A (MLL1) fusion protein MLL-AF9, CDX4 and MEN1 bind to the same regulatory region at the HOXA9 promoter in vivo, and MEN1 is needed for access of CDX4 to the HOXA9 promoter, likely through the establishment of H3K4me3 marks of active chromatin by the complex of MLL1 and MEN1 (Yan et al. 2006). Human CDX4 was shown to bind to promoter elements of the human HOXA10 gene, using pro-myeloid human cancer cell line U937 (Bei et al. 2011). |
