| text | Alpha-catenin (CTNNA1) associates with CDH1 indirectly, through interaction with CDH1-bound beta-catenin (CTNNB1) or gamma-catenin (JUP, also known as plakoglobin) (Aberle et al. 1994; Huber et al. 1997; Chitaev and Troyanovsky 1998: human recombinant CDH1 was used and endogenous catenins from human epithelial cell lines HaCat and A431). Human breast cancer cell line MDA-MB-468 is CTNNA1 deficient, and although it forms adherens junction-like structures, it is unable to form normal epithelial colonies. Interaction between CTNND1 and CDH1 in MDA-MB-468 cell line strengthens upon CTNNA1 reconstitution, implying intra-CDH1:catenin-complex interaction between CTNND1 and CTNNA1 that is abrogated in CTNND1 splicing isoforms that contain exon C, encoding amino acids 626-631 (Troyanovsky et al. 2011: human recombinant CTNND1 splicing isoforms, human recombinant CDH1 and human recombinant CTNNA1 were used). CTNNA1 was first reported to associate with complexes of CDH1, CTNNB1 and JUP in the mouse fibroblast cell line NIH3T3 (Ozawa et al. 1989), and the three catenins, CTNNA1, CTNNB1, and JUP, could also be pulled down from human cancer cell line HeLa with a recombinant mouse CDH1 (Ozawa et al. 1989). It is likely that CTNNA1 associates with CDH1-bound CTNNB1 or JUP during posttranslational processing of CDH1 in the endoplasmic reticulum (Zhu et al. 2001), but since CTNNA1 is not implicated to be directly involved in CDH1 posttranslational processing and trafficking, it is shown to form a complex with CDH1 at the plasma membrane. |
