| text | EZH2, a component of the PRC2 histone methyltransferase complex (the core complex is composed of EZH2, EED2, RBBP4 or RBBP7, and SUZ12) binds the promoter of the CDH1 (E-cadherin) gene. Binding of both endogenous EZH2 and EED2 to the endogenous CDH1 gene promoter was demonstrated by chromatin immunoprecipitation (ChIP) in human cancer cells. In response to TGF-beta stimulation, EED2 levels increase and promote increased occupancy of the CDH1 promoter by EZH2 (Oktyabri et al. 2014). SNAI1 (Snail) (Herranz et al. 2008; Tong et al. 2012) and/or SNAI2 (Slug) (Tien et al. 2015) may be involved in the recruitment of the PRC2 complex to the CDH1 gene promoter. Histone-lysine N-methyltransferase EHMT2 may facilitate the recruitment of the PRC2 complex to the CDH1 gene promoter (Pan et al. 2015). Secreted extracellular HSP90 (eHSP90) may promote recruitment of EZH2 to the CDH1 gene promoter (Nolan et al. 2015). MUC1 (Mucin 1) was also reported to target EZH2 to the CDH1 gene promoter (Rajabi et al. 2017). Several long non-coding RNAs (lncRNAs) were reported to direct PRC2 to CDH1 gene promoter, including HOTAIR (HOX transcript antisense RNA) (Wu et al. 2015; Song et al. 2019), LINC01133 (Zang et al. 2016), MALAT1 (metastasis-associated lung adenocarcinoma transcript 1) (Chen et al. 2020), SNHG1 (small nucleolar RNA host gene 1) (Xiang et al. 2020), and MRPL23-AS1 (MRPL21 antisense RNA 1) (Chen et al. 2020). |
