| text | Casitas-B-lineage lymphoma protein-b (CBLB), a RING finger E3 ubiquitin-protein ligase, was shown to inhibit NLRP3-dependent IL-1β release in human monocyte–derived and mouse bone marrow-derived macrophages (BMDM) (Tang J et al. 2020). In vivo, CBLB also prevented endotoxemia in mice induced by a sub-lethal dose of LPS in a caspase-11/NLRP3-dependent manner. The ubiquitin-associated (UBA) region of CBLB directly binds to the leucine-rich repeat (LRR) domain of NLRP3. CBLB is recruited to NLRP3 via its interaction with K63-linked polyubiquitin (Ub) chains attached by the E3-ligase activity of RING finger protein 125 (RNF125). CBLB then promotes K48-linked ubiquitination of NLRP3 at K496 inducing proteasome-mediated degradation of NLRP3 (Tang J et al. 2020). |
