| text | Casitas-B-lineage lymphoma protein-b (CBLB), a RING finger E3 ubiquitin-protein ligase, was shown to inhibit NLRP3-dependent IL-1β release in human monocyte–derived and mouse bone marrow-derived macrophages (BMDM) (Tang J et al. 2020). In vivo, CBLB also prevented endotoxemia in mice induced by a sub-lethal dose of LPS in a caspase-11/NLRP3-dependent manner. Immunoprecipitation assay showed interaction between endogenous NLRP3 and CBLB in mouse macrophages upon NLRP3 inflammasome activation. The NLRP3:CBLB association was also detected upon coexpression of tagged proteins in human embryonic kidney 293T (HEK293T) cells (Tang J et al. 2020). Mutagenesis analysis showed that the ubiquitin-associated (UBA) region of CBLB binds to the leucine-rich repeat (LRR) domain of NLRP3. CBLB is recruited to NLRP3 via its interaction with K63-linked polyubiquitin (Ub) chains. The E3-ligase activity of RING finger protein 125 (RNF125) was shown to conjugate K63-linked Ub chains to the NLRP3 LRR domain. CBLB then promotes K48-linked ubiquitination of NLRP3 at K496 inducing proteasome-mediated degradation of NLRP3 (Tang J et al. 2020). |
