textCDH11 intracellular domain associates with several catenin proteins. The juxtamembrane domain (JMD) of CDH11, located between amino acids 592 and 630, binds to CTNND1 (also known as p120 catenin or delta-catenin). The C terminal binding sequence (CBS) of CDH11, located between amino acids 687 and 745, binds to CTNNB1 (beta catenin) (Ortiz et al. 2015). The coordinates of JMD and CBS of human CDH11 were obtained from Kiener et al. 2006. The interaction of CDH11 with CTNNB1 is strong, as it is resistant to most detergents used to isolate CDH11 from the plasma membrane; the interaction of CDH11 with CTNND1 is weaker and more sensitive to detergent exposure (Lee et al. 2018).

CDH11 forms a complex with JUP (Junction Plakoglobin, also known as gamma catenin). In cow, rat and pig, JUP was shown to co-localize with CDH11, CTNNB1 and CTNND1 in the cortex adherens, a giant cell-cell adhesive complex formed by eye lens fiber cells (Straub et al. 2003). JUP co-immunoprecipitates with CDH11 (Lee et al. 2018). Based on studies with other cadherins, JUP and CTNND1 can simultaneously bind CDH11, while binding of CTNNB1 and JUP to CDH11 is mutually exclusive (Chitaev and Troyanovsky 1998; Zhurinsky et al. 2000; Miravet et al. 2003; Leonard et al. 2008).

Based on studies on N-cadherin (Wahl et al. 2003), it is likely that binding of catenins occurs in the endoplasmic reticulum, during trafficking of CDH11 to the plasma membrane.
