| text | Transcription of the CDH11 gene is governed by several transcription factors. ILF3 and HOXC8 transcription factors, stimulate CDH11 transcription both individually and cooperatively, through complex formation (Lei et al. 2005, Lei et al. 2006, Li et al. 2014, Zhang et al. 2017). ZEB2 stimulates CDH11 transcription in an SP1-dependent manner, which is thought to contribute to epithelial-to-mesenchymal transition (EMT) (Nam et al. 2014). HEYL directly stimulates CDH11 gene transcription. Both HEYL and CDH11 are frequently overexpressed in gastric cancer, with high HEYL levels correlating with poor prognosis (Liu et al. 2020). FOXF1 inhibits CDH11 gene transcription in both human and mouse fibroblasts. Direct repression of CDH11 gene transcription by FOXF1 has been demonstrated in mouse cells. FOXF1-mediated repression of CDH11 transcription, accompanied by FOXF1-mediated activation of CDH2 transcription, is thought to inhibit pulmonary fibrosis by preventing CDH2-CDH11 switch in myofibroblasts. FOXF1-mediated repression of CDH11 gene transcription is associated with the appearance of the repressive H3K27Me3 trimethylation mark on nucleosomes associated with the CDH11 gene promoter (Black et al. 2018). Based on a mouse study, the transcription factor BHLHE22 (also known as BHLHB5) recruits PRDM8, a histone methyltransferase, to the first intron of the CDH11 gene and directly represses CDH11 transcription. The repression of CDH11 expression by BHLHE22 and PRDM8 is important for the proper axon targeting of corticospinal motor neurons (Ross et al. 2012). During differentiation of human trophoblasts, TGF-beta-1 (TGFB1) positively regulates CDH11 gene transcription through upregulation of the SNA1 (Snail) transcription factor. While SNAI1 binding sites have been identified in the CDH11 promoter, direct binding has not been reported (Cheng et al. 2018). Positive regulation of CDH11 transcription by SNAI1 has also been reported in synovial fibroblasts (Chen et al. 2015; Cheng et al. 2015). |
