textAfter initial activation of Nkx2-5 and Gata4 by Mesp1, Gata4 binds the promoter of the Nkx2-5 gene and activates transcription (Lien et al. 1999, Reecy et al. 1999, Brown et al. 2004, He et al. 2011, He et al. 2014). Smad1:Smad4 complexes that are apparently activated by Bmp signaling also bind the Nkx2-5 promoter, recruit Gata4, and synergistically enhance transcription (Brown et al. 2004). In the second heart field (SHF) Mef2c mediates the binding of Nkx2-5 to its own promoter (Clark et al. 2013). Tbx20 binds the cardiac enhancer of the Nkx2-5 promoter and activates transcription (Takeuchi et al. 2005)
