textParamyxovirus V proteins target both IFIH1 (MDA5) and DHX58 (LGP2) helicase domains (Andrejeva J et al. 2004; Childs K et al. 2007; Rodriguez KR and Horvath CM 2013). Analysis of the interaction interface revealed the IFIH1 helicase C domain as necessary and sufficient for association with V proteins from human parainfluenza virus type 2, parainfluenza virus type 5 (PIV5), measles virus (MeV), mumps virus, Hendra virus, and Nipah virus (Parisien JP et al. 2009). Structural and biochemical studies showed that the viral V protein interaction disrupts ATP hydrolysis mediated by both IFIH1 and DHX58 (Parisien JP et al. 2009; Motz C et al. 2013). Expression of either MeV or PIV5 P/V protein antagonized IFIH1 (MDA5) signaling and also prevented DHX58 (LGP2)-mediated IFIH1 enhancement in human embryonic kidney 293 cells (Rodriguez KR and Horvath CM 2014). This Reactome event shows the ineraction of MeV P/V with both IFIH1 and DHX58.
