| text | FOXD3 is a negative regulator of melanocyte fate determination. FOXD3 is expressed in neural crest cells in a manner that depends on SNAIL2 and SOX9. SNAIL2 expression, in turn, depends on transcriptional regulation by neural border fate regulators ZIC1 and PAX3 (Plouhinec et al, 2014; Nitzan et al, 2013; Millet et al, 2013; Bae et al, 2014). These premigratory neural crest cells give rise to an early-migrating lineage that produces Schwann cell precursors and glial and neural cells and a later-migrating lineage that gives rise to the melanocytic lineage (reviewed in Mort et al, 2015; White and Zon, 2008). Expression of FOXD3 is maintained in the early-departing, ventrally-migrating cell population that gives rise to neural tissue, Schwann cells and glia, but is repressed prior to the delamination and migration of later-departing cells that travel dorsolaterally and give rise to the melanocyte lineage. The mechanisms of FOXD3 repression are not worked out in detail, but repression in zebrafish depends on HDAC1 (Ignatius et al, 2008). |
