| text | Casein kinase II (CK2) is a multifunctional serine/threonine kinase composed of two catalytic subunits (CK2α or α′) and two regulatory subunits (CK2β) (Meggio F et al. 2003). CK2 is involved in the immune regulation by phosphorylating antiviral innate immune response receptor RIG-I (DDX58) (Sun Z et al. 2011). Coimmunoprecipitation experiments showed that endogenous CK2 binds DDX58 in HEK293T cells. Similar results were found for ectopically expressed CK2 and DDX58 (Sun Z et al. 2011). Overexpression of CK2 inhibited DDX58-mediated antiviral signaling in human embryonic kidney 293T (HEK293T) cells and human umbilical vein endothelial cells (HUVEC). CK2 phosphorylates the C-terminal repressor domain (RD) of DDX58 at S854, S855 and T770 in the resting state of the cells, In contrast, viral infection led to dephosphorylation of DDX58 (Sun Z et al. 2011). CK2-mediated phosphorylation is believed to prevent DDX58 downstream signaling in the absence of viral RNA stimulation. Similar findings were reported in the studies with DDX58 phosphorylated in the N-terminal region at S8 and T170 (Wies E et al. 2013). Dephosphorylation of DDX58 is thought to be essential for the formation of DDX58 multimer (Sun Z et al. 2011). In addition, CK2 was reported to control the activation of components of DDX58 signaling pathway such as TBK1 and IRF3 in human and murine cells (Du M et al. 2015). This Reactome event describes CK2-mediated phosphorylation of DDX58 (RIG-1) at S854, S855 and T770. |
