textThe mitochondrial antiviral signaling protein (MAVS) is activated upon viral infection forming prion-like aggregates which are highly potent in activating downstream signaling pathways (Hou F et al. 2011; Xu H et al. 2014). The NOD-, LRR- and pyrin domain-containing protein 3 (NLRP3) was shown to physically interact with MAVS (Subramanian N et al. 2013; Park S et al. 2013). MAVS is thought to recruit NLRP3 to the outer mitochondrial membrane promoting the oligomerization of NLRP3 (Park S et al. 2013). NLRP3 oligomerizes via its nucleotide-binding and oligomerization domain (NACHT, also known as NOD) (Duncan JA et al. 2007). NOD of NLRP3 displays ATPase activity that is required for NLRP3 oligomerization (Duncan JA et al. 2007). The specific NLRP3 inflammasome inhibitor MCC950 targets the Walker B motif within the NOD domain of NLRP3 and blocks ATP hydrolysis inhibiting NLRP3 oligomerization and activation (Coll RC et al. 2019; Tapia-Abellán A et al. 2019). The oligomerization of NLRP3 is thought to serve as a platform to faclitate the assembly of apoptosis-associated speck-like protein containing a CARD (ASC or PYCARD) monomers into a prion-like polymer (speck).
