textBased on studies with NPM-ALK fusion proteins, ALK appears to signal through JAK3 in a manner that depends on JAK3-ALK binding (Zamo et al, 2002; Amin et al, 2003; Lai et al, 2005). When co-expressed in HEK293 cells, JAK3 binds to residues corresponding to 1058 to 1077 of the full length ALK receptor (Zamo et al, 2002). Although JAK3 often signals through recruitment and phosphorylation of STAT3, it is unclear whether STAT3 activation downstream of ALK depends on JAK3 (Zamo et al, 2002; Amin et al, 2003; Marzec et al, 2005; Han et al, 2006 a,b; Lai et al, 2005; Shi et al, 2006; reviewed in Turner and Alexander, 2006; Palmer et al, 2009).
