textThe tripartite efflux system MacA:MacB:TolC in enterobacteria confers resistance to macrolides, for example azithromycin. The system consists of a porin TolC trimer in the outer membrane, the ATPase inner membrane pump MacB, and a binding protein MacA that increases the pump's capacity and affinity to macrolides. The TolC trimer projects an α-helical tunnel across the periplasm, ending at the inner membrane transporter. The transport consists of two reactions, both catalyzed by the periplasm-spanning complex. In the first reaction catalyzed by the MacB subunit, one molecule of ATP is spent for the export of azithromycin out of the cytosol (Kobayashi et al. 2001; Pei et al., 2011; reviewed by Alav et al., 2021).
