| text | Ifenprodil is a phenylethanolamine-related compound that binds specifically to GluN1:GluN2B NMDA receptors. Ifenprodil shows the highest affinity for GluN1:GluN2B di-heterotetramers (Hatton and Paoletti 2005). One molecule of ifenprodil binds to each interface of GluN1 and GluN2B, interacting with N-terminal domains of GluN1 and GluN2B (Karakas et al. 2011, Tajima et al. 2016). Two molecules of ifenprodil bind to the GluN1:GluN2B di-heterotetramer (Hatton and Paoletti 2005). Ifenprodil acts as an allosteric inhibitor that stabilizes the closed conformation of the GluN1:GluN2B di-heterotetramer but does not prevent ligand binding to the NMDAR (Tajima et al. 2016). It is uncertain whether ifenprodil interferes with the release of the magnesium block. |
