commentFUNCTION The anti-Muellerian hormone (AMH) plays an important role in several reproductive functions (PubMed:11606457, PubMed:11861535, PubMed:1782869, PubMed:9435237). Anti-Muellerian hormone binds and activates AMHR2, its specific type-II receptor, that heterodimerizes with type-I receptors (ACVR1 and BMPR1A) to regulate target gene expression through downstream SMAD protein signal transduction (By similarity). Produced and secreted by Sertoli cells of the male fetus, anti-Muellerian hormone induces Muellerian duct regression during male fetal sexual differentiation (PubMed:1782869, PubMed:9435237). In female, it is produced by granulosa cells of the preantral and small antral follicles and acts as a negative regulator of the primordial to primary follicle transition and decreases FSH sensitivity of growing follicles (PubMed:11606457, PubMed:11861535). Also plays a role in Leydig cell differentiation and function (PubMed:1782869, PubMed:9435237).FUNCTION Plays an important role in several reproductive functions, including Muellerian duct regression during male fetal sexua,l differentiation and in the adult plays a role in Leydig cell differentiation and function (PubMed:1782869, PubMed:9435237). In female acts as a negative regulator of the primordial to primary follicle transition and decreases FSH sensitivity of growing follicles (PubMed:11606457, PubMed:11861535). Binds to its sole type II receptor, AMHR2 that recruits type I receptors ACVR1 and BMPR1A which subsequently activates the Smad pathway (By similarity).SUBUNIT Homodimer; disulfide-linked (By similarity). Anti-Muellerian hormone (AMH) is secreted as part of a non-covalent heterotetrameric complex consisting of AMH prodomain and anti-Muellerian hormone homodimers (By similarity). After proteolytic processing, the dimeric AMH prodomain remains non-covalently associated with the active homodimeric anti-Muellerian hormone, amplifying its biological activity (By similarity). The AMH prodomain homodimer is displaced upon binding of anti-Muellerian hormone to its receptor (By similarity).SUBCELLULAR LOCATION Expressed in Sertoli cells of fetal testes, and in testes just after birth, but absent in adult testes. In female, AMH is expressed after birth in the granulosa cells of the follicle.PTM The preproprotein is proteolytically processed to generate the AMH prodomain and the anti-Muellerian hormone (By similarity). Both homodimerize covalently, and homodimers associate to form a non-covalent heterotetrameric complex (By similarity).DISRUPTION PHENOTYPE Homozygous null mutant males have a complete male reproductive tract and functional sperm, but also uterus and oviducts (PubMed:7954809). Most are infertile due to female organs blocking sperm transfer. Females are fertile with enlarged ovaries and atypical follicles (PubMed:7954809). Furthemore in the absence of AMH, follicle growth is more sensitive to stimulation by FSH (PubMed:11606457).SIMILARITY Belongs to the TGF-beta family.
