textThe ERK orthologue, Rolled (RL), bound to the Mnk1/2 kinase orthologue LK6, phosphorylates it at Thr279 and Thr284 (inferred from Thr244 and Thr249 in mouse Mnk2) in the activation loop of the kinase catalytic domain. Thr424 (inferred from Thr379 in mouse Mnk2) C-terminal to the catalytic domain, is phosphorylated and these three residues are crucial in the activation of LK6. In addition, Ser1110 and Ser1113 (inferred from Ser431 and Ser434 in mouse Mnk2), believed to be involved in regulating the binding of RL to LK6, are phosphorylated. This results in the dissociation of LK6 to RL. It should be noted that the binding site on LK6 used by RL is also used by other proteins such as phosphatases, meaning that while RL occupies this site then LK6 can't be dephosphorylated.
