textExpression and function of the GP1BA:GP1BB:GP9:GP5 (GP1B-IX-V) complex is regulated by shedding of receptor ectodomains from the platelet surface (reviewed in Montague SJ et al., 2018; Wu J et al., 2020). The ectodomain of GP1BA (GP1B subunit α) is continuously cleaved by a disintegrin and metalloproteinase 17 (ADAM17, also known as tumor necrosis factor-alpha converting enzyme TACE) (Andrews RK et al., 2003; Bergmeier W et al., 2004; Gardiner EE et al., 2007). The cleavage site is located within the mechanosensory domain of GP1BA. The ADAM17-mediated cleavage of GP1BA releases a fragment known as glycocalicin (GP1BA(17-506)) (Andrews RK et al., 2003; Gardiner EE et al., 2007). Similarly, the ectodomain of GP5 (GPV) is cleaved releasing GP5 ectodomain (Rabie T et al., 2005; Gardiner EE et al., 2007). ADAM17 is the most important protease involved though other proteases such as ADAM10 may play a role (ColciaghiF et al., 2002, Gardiner EE et al., 2007).
