textThe NC1 domains of collagen types VII and X interact with integrin alpha2beta1. During endochondrial ossification, collagen type X is deposited in the hypertrophic zone of the growth plate. It interacts directly with chondrocytes via integrin alpha2beta1. Mutation in COL10A1 can cause disorders such as metaphyseal chondrodysplasia type Schmid (MCDS). Various missense mutations in COL10A1 are classified into three classes based on their functional consequence. Class I mutations disrupt the hydrophobic core of COL10A1 and class II mutations can fold but perturb trimer assembly, both of which cannot form collagen structures. However, class III mutations are capable of folding and forming abnormal fibril structures (Marks D S et al. 1999, Bogin O et al. 2002).
