textInterleukin-18 (IL18) is suggested to be cleaved by Caspase-1 active tetramer complex (Ghayur et al. 1997). This cleavage is necessary to activate IL18 and to generate an inflammatory response capable of eradicating some pathogens, as Shigella, effectively (Sansonetti et al. 2000). Note that Asp has been identified as the cleavage site in IL18 for Caspase-1 (Tan et al. 1998). In addition, IL18 induces Interferon gamma (IFNG) and activates a Th1 response. Invasion of macrophages by Shigella directly activates CASP1, which triggers an acute inflammation by proteolytically activating IL1beta and IL18. The active tetramer of Caspase-1 (CASP1) contains at least two dimers formed by subunits 120-197 and 317-404 (Thornberry et al. 1992). Gaggero et al. 2004 also suggest that other caspases can activate IL18.
