textActivated STAT complexes induced by Interleukin-9 (IL9) were found to contain Signal transducer and activator of transcription 1-alpha/beta (STAT1), Signal transducer and activator of transcription 3 (STAT3), and Signal transducer and activator of transcription 5A or 5B (STAT5A or STAT5B) transcription factors. STAT1:STAT3 heterodimer, and 2xp-STAT1 or 2x(STAT5A,STAT5B) homodimers binds to DNA. Interleukin-9 (IL9) has shown to activate STAT by mean of using a 32P-labeled oligonucleotide, a motif which is known to avidly bind STAT factors. Wild-type BW5147 cells were treated or not with mouse IL9. IL9-induces DNA-binding complex , and this reaction can be selectively inhibited by a cold competitor excess (data not shown) (Demoulin et al. 1996).
